其实个人认为没有什么大的不同.
第一个应该是作“休息三天”之意,原句应该是 I had ~之类的.
第二个,算是词组吧,就是“三天的休息”之意.
第三个,真的搞不懂,为什么有了-后面day还用s呢?无能为力.不对吧.
第四个,是一个名词词组吧.“一个三天的休假”,用法应该和第二个差不多.
本人不才,英语不好,略发拙见,仅供参考.

可以有复数,但必须是 days off；
可以用 have 也可用 take 当动词.
例如：

I am going to take a few days off next month.（下个月我打算休几天假.）
