That entire portion of the Mexican anti-drug campaign is a fraud, as Salazar is wiping out one cartel because he has aligned with another for profit. Javier "s partner, Sanchez, attempts to sell the information of Salazar"s true affiliation to the DEA, but is killed for his betrayal. Javier , who can no longer stomach working for Salazar, decides to make a deal with the DEA. In exchange for his testimony, Javier requests electricity in his neighborhood, so the kids can play baseball at night rather than be tempted by street gangs and crime. Wakefield storyline Meanwhile, Robert Wakefield (Douglas), a conservative Ohio judge, is appointed to head the President"s Office of National Drug Control, taking on the title drug czar. Robert is warned by his predecessor (Brolin) and several influential politicians that the War on Drugs is unwinnable. His daughter, Caroline (Christensen), an honors student, has been using cocaine and develops a drug addiction after her boyfriend Seth (Grace) introduces her to smoking it. Caroline and Seth are arrested when a fellow student overdoses on drugs at a party. As Robert and his wife Barbara (Irving), struggle to deal with the problem he discovers that she has known about their daughter"s involvement with drugs for over six months. 这涉及到抗体识别抗原的原理和抗体的结构。比如你要检测的蛋白为A，它有a, b, c, d 四个表位(epitope)，可以被抗体特异性认识。制作针对A的抗体是将蛋白A的一段，比如包含b+c段，打入兔子（最常见）或其他动物（山羊goat，大鼠rat，鸡chicken，仓鼠hamster），由于这是外来入侵物，动物自身的免疫系统会生产针对它的抗体来清除这个异物，而产生的抗体有些会识别b位点，有些则识别c位点。之所不用完整的蛋白，是因为全长通常会太大。然后收集被免疫了的动物的血清，这个含有针对蛋白A的的多克隆抗体（因为是识别c+d的混合抗体）就称为抗血清（antiserum)。有的产品卖的直接就是抗血清，有的则再一步将抗体纯化出来。这种多克隆抗体一般会便宜一些。而通过体外刺激小鼠的B细胞和肿瘤细胞的杂合体，分享单克隆进行培养，可以产生出只针对一个表位b的单克隆抗体（monoclonal antibody），这种抗体的特异性更高，但价格也更贵。但具体到用单抗还是用多抗，则要具体情况具体分析。比如你要测的蛋白浓度很低，用多抗检测到的可能性会比用单抗高。又或者待测蛋白会被加工切割成两段，要想两段都检测到，显然应该用针对两段表位的多抗。由此可见，一抗都是特异性针对你要检测的蛋白，你要测A，他要测B，我要测C,所以产量就不可能太大，因为需求量没有那么大。而二抗是个什么情况呢？首先要了解抗体的结构。抗体结构就像一个Y字形，两个丫的部分称为可变区（Fab)，正是这个区域特异性地识别抗原的表位。就像一把钥匙配一把锁一样，表位b只被含有b"可变区的抗体识别。而抗体Y字形下面的主干部分称为恒定区(Fc)，所有人的抗体的Fc都是一样的，不同的只是Fab区，可以是b"，可以是c"。所有鼠的抗体Fc也都是一样，所有山头的Fc也都是一样。所以制二抗，就是用一抗的Fc区去免疫动物，方法同制作一抗。注意如果一抗是在兔子上制作的，那么制二抗时就不用再打兔子了，因为兔子不会对句子的Fc产生免疫反应，不然就会产生自身免疫疾病，自己产生抗体攻击自己。有一部分不幸的人就患有自身免疫疾病。话说回来，从兔子中抽出的任意抗体，不一定要一抗（因为所有的Fc都一样），制作只含有Fc段的片段，再打到山羊中，从而获得了针对兔Fc的抗体。这个抗体能够识别所有来自于兔子体内的抗体，不论它们的Fab段是什么。也就是说二抗的适用范围远远大于一抗，因为一抗是一对一（Fab对表位），二抗是一对多（动物二Fab对动物一Fc）。所以关键在于不是二抗抗一抗不特异，而是二抗抗一抗的恒定区。

根据南传上座部佛教，白骨观的说法不是很准确，佛陀教导的四十种禅定方法中，十不净的骸骨相比较接近白骨观的说法。因此，白骨观应该是不净观的内容之一。不净观可分为外在尸体的不净观与内在三十二身分的不净观，这两种不净观都能去除贪欲。其中三十二身分是观身体三十二个组成部分为不净。外在尸体的不净观是观尸体的膨胀相、青瘀相、脓烂相、断坏相、食残相、散乱相、斩斫离散相、血涂相、骸骨相、虫聚相为十不净。------前九种也可以称作九种墓园观、九种坟场观。详细修法如下：三十二身分，佛陀在《大念处经》教导：「再者，诸比丘，比丘思惟这个身体，从脚掌思惟上来及从头发思惟下去，这个身体由皮肤所包裹，并且充满许多不净之物：『在这个身体当中有头发、身毛、指甲、牙齿、皮肤、肌肉、筋腱、骨骼、骨髓、肾脏、心脏、肝脏、肋膜、脾脏、肺脏、肠、肠间膜、胃中物、粪便、胆汁、痰、脓、血、汗、脂肪、泪液、油脂、唾液、鼻涕、关节滑液、尿液。』」像经文所教导的这样，以正念观照身体中各种不净物的修行方法称为不净观。这种修行方法是外道所没有的，只在佛陀出现于世间时才有。这种观身不净的正念能带来下列的利益：一、 大悚惧（mahasamvega）；二、 大瑜伽安稳（mahayogakkhema）；三、 大正念与正知（mahasatisampajabba）；四、 获得智见（banadassanapatilabha）；五、 当下的安乐住处（ditthadhammasukhavihara）；六、 现证智慧与解脱之果（vijjavimuttiphala- sacchikiriya）：即证悟阿罗汉道果。此法门有七种善巧的学习方法：一、 反复地念诵三十二身分（的名称）（vacasa）；二、 反复地记忆三十二身分（manasa）；三、 依照颜色来确定头发等各身分（vannato）；四、 依照形状来确定各身分（santhanato）；五、 依照其位置在肚脐的上方或下方来确定各身分（disato）；六、 依照所占有的部位来确定各身分（okasato）；七、 依照与其它部份划分的界限来确定各身分（paricchedato）。此法门有十种善巧的修行方法：一、 次第地观照（anupubbato）；二、 不要观照得太快（natisighato）；三、 不要观照得太慢（natisanikato）；四、 排除散乱（vikkhepa-patibahanato）；五、 超越概念（pannattisamatikkamanato）；六、 逐渐去除不清晰的身分（anupubbamubcanato）；七、 观照能导致安止的身分（appanato）；八、 九、十、依照《增上心经》（Adhicitta Sutta）、《清凉经》（Sitibhava Sutta）与《觉支善巧经》（Bojjhavgakosalla Sutta）的教导来修行。九种坟场观佛陀在《大念处经》以九种坟场观来解释身念处如下：「再者，诸比丘，比丘在坟场中见到死后经过一天、二天或三天的被丢弃尸体，那尸体肿胀、变色、腐烂。他拿自己的身体来和那具尸体作比较，内心思惟着：『确实地，我的身体也具有同样的本质，它将会变成那样，如此的下场是无法避免的。』」【二】「再者，诸比丘，比丘在坟场中见到被丢弃的尸体，被乌鸦、老鹰、秃鹰、苍鹭、狗、老虎、豹、豺狼或各种虫所噉食。【三】比丘在坟场中见到被丢弃的尸体，已经变成只剩下（一些）血肉附着的一具骸骨，依靠筋腱而连结在一起。【四】比丘在坟场中见到被丢弃的尸体，已经变成一具没有肉而只有血迹漫涂的骸骨，依靠筋腱而连结在一起。【五】比丘在坟场中见到被丢弃的尸体，已经变成一具没有血肉的骸骨，依靠筋腱而连结在一起。【六】比丘在坟场中见到被丢弃的尸体，已经变成分散在各处的骨头：这里一块手骨、那里一块脚骨、这里一块脚踝骨，那里一块小腿骨、这里一块大腿骨、那里一块髋骨、这里一块肋骨、那里一块背骨、这里一块胸骨、那里一块颈椎骨、这里一块下颚骨、那里一块牙龈骨、这里是头盖骨。【七】比丘在坟场中见到被丢弃的尸体，已经变成贝壳色的白骨。【八】比丘在坟场中见到被丢弃的尸体，已变成经过一年以上的一堆骨头。【九】再者，诸比丘，比丘在坟场中见到被丢弃的尸体，骨头已经腐朽成骨粉。他拿自己的身体来和那具尸体作比较，内心思惟着：『确实地，我的身体也具有同样的本质，它将会变成那样，如此的下场是无法避免的。』」虫观：在智慧之光的帮助下，去照见存在自己身体内的许多虫，见到之后就观照你的身体为不净。为什么呢？因为这些虫在你的身体里交配、生病、排放大小便、死亡，因此你的身体就是它们的家、医院、厕所、坟场。也必须观照外在他人的身体由于充满各种虫因此是不净的，如此交替观照内外身体的不净。满意之后就对那些虫修行四界分别观。见到色聚之后就分析色聚里的究竟色法，然后观照它们为无常、苦、无我、不净。

1.多普勒超声目前首选的无创性颈动脉检查手段，广泛应用于颈动脉硬化病变的筛查及随访。不仅可以显示斑块的部位和大小、管腔狭窄部位和严重程度，还能进行血流动力学测定，并可对斑块进行形态学评价。彩色多普勒超声通过检测颈动脉内膜中层厚度（IMT）来确定是否有动脉粥样硬化斑块形成。正常IMT应小于1.0mm，IMT在1.0～1.2mm间为内膜增厚，大于1.2mm为斑块形成。超声下根据形态和回声特点，斑块可分为①低回声脂质性软斑；②中等回声富含胶原组织的纤维性扁平斑块；③强回声伴声影的钙化性硬斑块；④回声强弱不等的溃疡性混合型斑块。其中，软斑、扁平斑和混合斑属于不稳定斑块，是引起缺血性脑卒中的重要原因之一。2.经颅多普勒超声（TCD）另一项无创检查手段，可显示颅内外动脉的狭窄部位、程度、血流速度、血流方向及是否有侧支循环开放等，经常与多普勒超声联合应用于颈动脉狭窄的诊断及术后评估。3.CT血管造影（CTA）CTA对颈动脉狭窄的判断准确性高于多普勒超声，但对斑块的形态学显示欠佳，目前广泛应用于狭窄的诊断，可作为术前诊断和制定治疗方案的重要依据。4.核磁血管造影（MRA）可清晰显示颈动脉及其分支的三维形态和结构，并重建颅内动脉影像，对狭窄程度较重的病变判断敏感性高，但价格相对较高，体内有金属植入物（如金属假牙、起搏器或金属假体等）者禁行此检查。5.数字减影血管造影（DSA）DSA是诊断颈动脉狭窄的金标准，可详细评价病变的部位、范围、程度以及侧支形成情况，但因属有创操作、价格昂贵、风险较高，临床上很少单纯用于检查。
