　　摘　要：人的HTRIE基因是五羟色胺受体(HTR)家族中的一员，这个家族里面的基因都和精神分裂症、抑郁症以及人的自杀活动有关，但是对于其具体的作用机制目前研究不多，采用实时定量PCR方法分析了HTRIE基因在各个组织和不同细胞系中的表达情况，亚细胞定位发现HTRIE位于细胞膜上，萤光素酶实验分析确定HTRIE可以抑制原癌基因erbB2启动子的转录活性，这些研究结果为进一步研究HTRIE基因在疾病发生中的作用奠定了一定的基础。 　　关键词：HTRIE；表达谱；实时定量PCR：转录活性 　　中图分类号：Q7　文献标识码：A　文章编号：1007-7847(2007)04-0311-05 　　 　　人类的五羟色胺受体可以分为7种类型，即5-HTRI～7，这7种类型又可以分为14种亚型，分别为：HTR1A，HTR1B，HRTIC，HTRID，HTRIE，HTI1F，HTR2A，HTR2B，HTR2C，HTR3A，HTR4，HTR5A，HTR6，HTR7，有研芋云表明HTR7和HTR4都可以激活转录因子固醇反应元件(serumresponse element，SRE)的转录活性，在NIE-115细胞系中过表达5-HT4(a)可以使其变圆，在海马神经元中，HTR7有使神经轴突变长的功能，HTRlB能够和S100A10，Pll(annexin 2 lightchain)蛋白质发生相互作用，而且发现HTRlB对下游ERK1/2的Thr202/TYr204磷酸化具有调控作用，而且可以通过和pll相互作用抑制神经冲动的传导，人HTRIE基因位于染色体的6q14-q15，并且含有7个跨膜结构域，因此属于G-蛋白偶联受体家族，目前发现此蛋白质在背根神经中枢有表达，并且具有抑制cAMP的形成，而且在去甲肾上腺素、肾上腺素、组氨、多巴胺和褪黑激素的刺激下，其蛋白质的表达情况没有任何变化，研究发现在阻断剂methiothepin的刺激下，HTRlE抑制cAMP的形成作用消失，有研究表明，五羟色胺诱导钙离子释放是通过激活HTR1E来实现的，而且发现HTtR1E可以调节细胞因子IL-6和CXCL8/IL8的释放，因此HTR1E可能在激活各种细胞信号途径以及调节细胞因子的释放起到了很重要的作用，我们通过对HTRIE基因在各个组织和细胞系中的表达情况以及亚细胞定位分析，为以后对HTRIE功能的研究打下基础。 　　 　　1　材料与方法 　　 　　1．1　材料 　　真核表达载体pEGFP-C3和含有erb B2启动子的报告基因pTAL-luc-erbB2购自Clontech公司，pMDl8-T载体、不含erbB2启动子区域的报告基因的载体pTAL-luc、各种限制性内切酶、T4DNA连接酶、T4聚合酶及反转录试剂盒购自TaKaRa，LipofectamineTM 2000转染试剂盒，总RNA提取的TRIZOL试剂购自Invitrogen，luciferase assay试剂盒购自Promega，SYBRGreen I荧光染料实时定量PCR试剂盒购自Applied Bio systems公司，倒置荧光显微镜为Zeiss产品，PE全自动荧光定量PCR仪为美国ABI 7900产品，荧光分光光度计TD-20/20为美国产品，T47D、HeLa、HepG2、Hep3B、SW480、MCF7等14种细胞系以及人的各种组织由实验室保存。 　　 　　1．2　方法 　　1．2．1　细胞培养、总RNA的提取、cDNA的合成T47D、HeLa、HepG2、Hep3B、SW480、MCF7等细胞在37℃，5％CO2，相对湿度95％条件下，用完全培养液DMEM(补加10％新生牛血清、2mol/L谷氨酰胺和100mg/L青霉素和100mg/L链霉素)培养，24h以后用PBS将细胞洗两次，然后用细胞刮收集细胞，各种细胞系和组织按照Invitrogen公司试剂盒提供的方法进行，用反转录试剂盒合成各细胞系和组织的cDNA片段。 　　1．2．2　目的基因的扩增以及重组载体的构建 　　从人大脑组织中提取RNA，然后以反转录的cDNA为模板，以HTRIE引物对(sense primer：5′-CTCGAGATGAACATCACAAACTGTACCAC-3′，添加Xho I位点anti-sense primer 5′-GGTACCCTAAGTATGCTCTCGGCATCT-3′，添加gpn I)克隆人HTRlE编码序列(1098bp)，然后亚克隆到pMDl8-T载体中，用Xho I/Kpn I酶切鉴定正确后，用Xho I/Kpn I酶切将目的片段切下，再亚克隆到同样酶切处理的pEGFP-C3表达载体中，酶切验证后测序分析，以确保读码框和序列的正确性。 　　1．2．3　实时定量PCR 　　用Trizol抽提各种细胞系的细胞总RNA，取总RNA 2μg用SuperscriptⅡ逆转录试剂盒进行逆转录反应，再以逆转录反应液2μL作为模板，对TNFAIP l基因的表达采用实时荧光定量PCR检测，定量引物采用Primer Express 3.0软件设计(sense primer：5′-TCCACCAG CCTGCCAACTAC-3′anti-sense primer 5′-CCGTCCTCq"TCCTGGCGTAT-3′)；反应体系为：1×SYBR Green PCRMaster Mix，50mmol/L sense primer，50 mmol/Lanti-sense primer，100ng模板进行，反应条件为：95℃变性15s、60℃复性60s，72℃延伸60s，共40个循环，以β-actin作为内参照，用去离子水取代模板作阴性对照，用美国PE Biosystems公司提供的SDS2软件进行数据处理，以RO值为纵坐标做直方图，比较表达差异，每个样品重复3次。 　　1．2．4　HTRIE亚细胞定位 　　用完全培养液DMEM培养Hep3B细胞长到70％～80％时，将pEGFP-C3-HTRIE转染到Hep3B细胞，细胞培养24h后，用PBS洗细胞一次，然后用4％的多聚甲醛室温固定10min，PBS洗两次，随后将50％的丙酮和50％甲醇混合液加入到细胞中室温放置30s后，PBS洗两次，然后加入终浓度为0.5mg/L的Hochest染色液，室温放置30min，用PBS洗两次后在荧光显微镜下观察细胞。 　　1．2．5　萤光素酶实验检测HTR1E对erbB2转录活性的影响 　　用完全培养基DMEM培养HEK293Fr细胞，等到细胞长到70％～80％时，将pTAL-luc-erbB2与pEGFP-C3-HTRIZ，pTAL-luc与pEGFP-C3-HTRIE，pEGFP-C3与pTAL-luc-erbB2，pTAL- luc与pEGFP-C3分别共转染到培养好的HEK293Fr细胞中，每组重复3次，每孔用LacZ共转染到细胞中来平衡用量，转染24h后，将细胞用PBS洗两次，裂解细胞抽提蛋白，测定各组分蛋白的荧光强度。 　　 　　2 结果 　　 　　2．1 细胞和组织的总RNA的提取及构建真核表达载体的成功 　　将细胞和组织中提取的RNA各取2μL进行琼脂糖电泳，可以观察到清晰的28S、18S、5S条带，RNA的提取效果理想，然后检测各RNA的A260/A280值，其A260/A280值都在1.85左右，说明提取的RNA有着较高的浓度(图1)，反转录后，克隆的人HTRIE基因片段在约1000bp处获得单一目的条带，与克隆基因1098bp的大小基本一致(图2)，克隆基因亚克隆到真核表达载体pEGPF-C3中，Xho I/xpn I酶切验证得到一条约1000 bp的条带(图3)，测序结果进一步说明HTRIE基因亚克隆到表达载体中，而且读码框正确。 　　 　　2．2 HTRIE在各个组织和细胞系中表达分析 　　用RT-PCR检测HTR1E在常见的细胞系和组织中的表达情况(图4)，结果显示HTRIE基因在细胞中的表达情况为：人的乳腺癌细胞T47D和纤维素瘤细胞HTl080-Tetoff表达量最低，而在人的肝癌细胞HepG2表达量最高，在所有检测的组织中都有较高的表达，但是在肝脏组织表达量最高，在脾脏组织表达量最低，在实验过程中，目的基因和内参β-actin在琼脂糖电泳后都只有单一的一条带，说明两个基因都是特异性的扩增，实验没有污染(结果没有显示)。 　　 　　2．3　HTRIE定位于细胞膜上 　　在Hep3B细胞中转染pEGFP-C3-HTR1E，经过Hocheset染色后，然后在荧光显微镜下观察细胞核和目的基因的分布情况，结果发现HTR1E定位于细胞膜上(图5)。 　　　 　　 　　2．4 HTRIE可以抑制原癌基因erbB2的转录活性 　　将p7AL-luc-erbB2与pEGFP-C3-HTRIE,pTAL-luc与pEGFP-C3-HTRIE，空载体pEGFP-C3与pTAL-luc-erbB2，pTAL-luc与空载体pEGFP-C3分别共转染到HEK293FT细胞中，每组重复3次，转染24h后，测定报告基因的荧光值，结果发现在HEK293Fr细胞中过表达H7R1E后，报告基因erbB2的荧光值明显下降，没有erbB2启动子的报告基因的pTAL-luc在过表达H7R1E后荧光值没有明显的下降的趋势，这说明HTRlE对原癌基因erbB2的转录有抑制作用(图6)。 　　 　　3 讨论 　　 　　原癌基因ERBb2是表皮生长因子受体(EGFR)家族中的一员，研究发现，在小鼠中敲除erbB2基因11d后小鼠死去，并且其运动神经和颅神经嵴衍生感官神经节的发育也受到了严重的影响EGFR(erbBl)，erbB3和erbB4 3个基因都需要与配体的结合才能够被激活，但是erbB2的激活并不需要与配体的结合，这意味着其他的信号分子也可以通过erbB2来传递信号，目前对与erbB2致癌的研究主要集中在细胞水平，并已经证实了erbB2可以通过G-蛋白偶联受体的信号通路来激活，在许多类型癌症中erbB2都出现过表达的情况，所以在治疗癌症时，erbB2也成为药物治疗的靶标，我们通过对HTREI的亚细胞定位发现，作为G-蛋白偶联受体的HTRIE定位于细胞膜上，而且发现此基因在各个组织和细胞中都有表达，并可以抑制erbB2的启动子的转录活性，这也就意味着HTRIE基因可能在癌症的发生中扮演了重要角色，为以后HTRIE功能的研究打下了基础。 　　 　　作者简介：吴艳阳(1981―)，男，湖南张家界人，硕士研究生，主要从事转录因子调控机理的研究；张健(1963―)，男，湖南长沙县人，湖南师范大学教授，通讯作者，主要从事转录因子调控机理的研究。
